PDRN is earning a fast-growing reputation in the consumer press as skincare’s latest DNA-powered discovery. The current hype doesn’t tell you much about whether it’s worth putting in your formulation, though, or what you need to know before you do.Â
PDRN, or Polydeoxyribonucleotide, is a short-fragment DNA ingredient that’s working its way into many topical skin and scalp products. Â
This blog explains what PDRN is, how it works, the difference between animal and plant-derived versions, and where it fits into a product you’re developing.Â
What is PDRN?
PDRN is a low molecular weight compound that’s derived by breaking down full DNA strands into short, biologically active fragments that skin cells can bind to and reuse.Â
Traditionally, PDRN was extracted from the sperm cells of oily fish like chum salmon and rainbow trout. The DNA was purified, sterilised and fragmented to the right molecular weight for use. Â
More recently, plant-based PDRN has entered the market as an alternative source, which is a useful development if you’re formulating for a vegan, halal or kosher-conscious customer base, or want to sidestep the supply and safety questions that come with sourcing animal-based products. We’ll come back to that trade-off later, because it’s one of the more important decisions you’ll make when sourcing your PDRN.Â
How PDRN works on the skin and scalp
PDRN’s effect on skin comes down to two related mechanisms which cause it to behave differently to a lot of other actives you may already be using.Â
PDRN interacts with the A2a receptors found on the surface of key skin cells. When the skin’s enzymes break PDRN down, it releases adenosine, which triggers a biochemical cascade linked to reduced inflammation, increased growth factor activity and improved blood flow to the area. This is a signalling effect. PDRN is essentially telling skin cells to switch into repair mode.Â
The second mechanism works through what’s known as the ‘salvage pathway’. When the skin is damaged, cells often lack the energy, specifically ATP, needed to build new nucleotides from scratch, which slows down repair. PDRN provides ready-made nucleotide fragments that cells can reuse directly, bypassing that energy bottleneck and allowing healing and regeneration to happen faster than it otherwise would.Â
Together, these mechanisms translate into some useful downstream effects for your formulation. They support collagen and hyaluronic acid synthesis, a calmer inflammatory response, and better maintenance of the skin’s collagen, elastin and cross-linking enzymes that keep tissue firm and elastic. The same effect is also relevant to scalp health. Chronic low-grade inflammation is one of the drivers behind premature shedding. PDRN helps calm that inflammatory signalling, giving it a viable role in scalp and hair formulations, not just in facial skincare.Â
Animal-derived vs plant-derivedÂ
Animal-derived PDRN is the original and most established version of the ingredient, with the longest track record and the widest availability. But it comes with trade-offs you’ll need to account for. It excludes use in vegan, vegetarian and, often, halal or kosher products. Sourcing it depends on fishing or farming supply chains, which brings the same kind of exposure to regional and seasonal disruption that you’d see with any animal-derived raw material. And because it’s an animal by-product, there’s a theoretical risk of contamination, which typically means more intensive purification during processing.Â
Plant-derived PDRN addresses most of these concerns. It’s vegan by nature and animal-free, sourced from botanical material rather than fish, which removes the ethical issues and the contamination risk associated with animal tissue. The purification process tends to be gentler, too. It doesn’t need the harsh solvents often used to process animal-derived material to the required purity.Â
However, plant-derived PDRN typically produces shorter DNA fragments than the animal-derived version, in the region of 50 to 500 base pairs compared with up to 2,000 base pairs from fish sources. Practically, this will shape what you can put on your ingredient declaration, and which markets and consumer segments you can target. Â
Where PDRN fits in a formulation
PDRN turns up across a range of topical skin and scalp care products, from serums and creams to masks and scalp treatments. The format you choose can affect how your finished product performs and feels when used. Â
It’s a hydrophilic ingredient that disperses cleanly in water-based systems, which means you won’t need specialist equipment to work it into your formulation. It also layers well alongside the humectants and emollients you’d typically build a hydrating product around.Â
Most of the benefits from a topical PDRN-based product come from its hydrating, calming and barrier-supporting effects, alongside its role in supporting the skin’s own collagen and hyaluronic acid production. That’s a useful set of properties to build your formulation around, whether you’re developing a hydrating serum, a soothing post-treatment product or a scalp formulation aimed at calming inflammation and supporting healthier hair growth.Â
On the practical side, PDRN typically appears at low use levels, often around 1%, which makes it straightforward to work into serums, balms and lightweight lotions without disrupting the rest of your formulation. Heat stability is the main processing constraint to plan around. PDRN’s structure is at risk of degrading above roughly 40°C, so it needs to be added at a cooler stage of your process rather than exposed to prolonged heat during manufacture. That’s a standard consideration if you’re used to working with heat-sensitive actives, but it’s worth flagging early to your production team so it’s built into the process from the start rather than discovered at scale-up.Â
PDRN can also complement the actives you’re likely already using. It pairs well with hydrators like hyaluronic acid, and with other regenerative or barrier-supporting ingredients, giving you scope to build a formulation around it instead of treating it as a standalone hero ingredient. Â
